I am sure that my husband is having an affair. After 27 years of marriage and four children, if I file for divorce, can I ask for half of our assets, including anything he might inherit from his recently deceased Dad's estate?

I am sorry to hear that you suspect infidelity. If you fiel for divorce, you my request an even split of marital assets. But, inheritences are nto alway streated as marital property, so the inheritence may nto be split. But, what he gets form his Dad's estte may be considered in establsihing a support or division of property Order.
